DBC is the amount of target molecule that binds to a chromatography resin under active hydrodynamic flow before significant breakthrough (typically 5% or 10% of inlet feed concentration) occurs.
Why is Residence Time critical for DBC?
Protein mass transfer into porous resin beads is diffusion-limited. Increasing residence time (e.g., from 2 to 6 minutes) increases DBC until reaching the static equilibrium capacity.
Why subtract the void volume V₀?
Void volume accounts for liquid holdup in upstream tubing and column interstitial volume before the feed front actually passes through the resin bed.
