Traditional Japanese Natto & Bacillus Subtilis Microbiology
Natto is created through the fermentation of whole soybeans by Bacillus subtilis var. natto, an aerobic, spore-forming, heat-resistant bacterium. During incubation, the bacteria secrete extracellular proteolytic enzymes that hydrolyze soy proteins into free amino acids (glutamates) and synthesize long chains of poly-γ-glutamic acid (γ-PGA) and fructan, creating natto's signature viscous, sticky, stringy mucilage and potent cardioprotective enzyme nattokinase.
The Three Non-Negotiable Rules of Natto Making
- Extreme Tenderness: Steamed beans must be soft enough to easily crush between your thumb and pinky finger without resistance. Firm beans cannot be penetrated by Bacillus enzymes.
- Shallow Bed & Oxygen Exchange: Bacillus subtilis is an obligate aerobe. Beans must be spread in a shallow pan no deeper than 2 to 3 beans deep (≤ 1 inch / 2.5 cm) and covered with perforated plastic wrap to allow oxygen flow while retaining humidity. Deep beans will suffocate and rot.
- 24-Hour Refrigerator Conditioning: Freshly incubated natto has a harsh ammonia odor. Aging in the refrigerator for 24 to 48 hours allows the ammonia to dissipate and allows γ-PGA stringiness and rich umami to peak.
Frequently Asked Questions
Why must the spore inoculant be added while the beans are still steaming hot?
Unlike yeasts and molds which are killed by heat, Bacillus subtilis endospores are heat-resistant up to boiling temperatures. Inoculating while beans are 80°C to 90°C (176°F - 194°F) heat-shocks the dormant spores into rapid germination while instantly killing any stray wild bacteria or mold spores.
Why do smaller soybeans make superior natto?
Micro or small soybeans (Kotsubu) have a much higher surface-area-to-volume ratio than large soybeans. Since Bacillus subtilis grows entirely on the exterior bean surface, smaller beans yield significantly more sticky mucilage (neba-neba) and nattokinase per bite.
How long can homemade natto be stored?
Natto keeps fresh in the refrigerator for 1 to 2 weeks. For long-term storage, portion into small airtight containers or freezer bags and freeze for up to 6 to 12 months; the beneficial bacteria and nattokinase survive freezing undamaged.
